    Topical Anti-Inflammatory and Analgesic Activities of Citrullus colocynthis Extract Cream in Rats.

    Background and objectives:Citrullus colocynthis (CC), known as bitter apple, is used to treat diabetes in Iranian traditional medicine. The aim of this study is to evaluate the anti-inflammatory and analgesic activities of CC cream in rats. Materials and Methods: The carrageenan-induced edema in a rat hind paw was carried out to evaluate the topical anti-inflammatory effect of the CC fruit extract cream (2⁻8%) and the tissue levels of IL-6 and TNF-α were estimated by using a commercial ELISA kit. The topical antinociceptive activity of CC cream (2⁻8%) was evaluated in the rat formalin test. To determine the role of opioid receptors in the local antinociceptive effect of the CC cream, naloxone (20 μg/paw, i.pl.), a non-selective opioid antagonist, was used. Results: The results showed that the CC cream (2⁻8%) dose-dependently reduced the carrageenan-induced paw edema and reversed the changes in the level of TNF-α and IL-6 due to carrageenan-induced edema (p < 0.01). The anti-inflammatory effect of CC cream 8% was comparable to that of hydrocortisone ointment 1%. Furthermore, the application of CC cream (2⁻8%) dose-dependently inhibited both first and second phases of the formalin test (p < 0.05). The antinociceptive effect of the CC cream (8%) was comparable to that of methyl salicylate cream 30%. Moreover, the administration of naloxone significantly reversed the topical antinociceptive effect of the CC cream (p < 0.05). Conclusions: For the first time, this study indicated that the topical application of CC cream possesses significant anti-inflammatory and antinociceptive activities in animal models, which were probably mediated by opioid receptors and the suppression of pro-inflammatory cytokines (TNF-α and IL-6). Thus, the CC cream can be used to treat inflammatory pain and inflammatory diseases

    Particulate Matter Inhalation Exposure Chambers and Parameters Affecting Their Performance: A Systematic Review Study

    Exposure to inhalation aerosols and particulate matter (PM) in different concentrations can increase the risk of respiratory, cardiovascular, and other related diseases. The inhalation exposure studies are implemented to assess the biological effects of these hazardous agents in human or animal models, in whole-body (WB) or nose/head-only conditions. Several factors can affect the performance of the inhalation exposure chambers and if left uncontrolled, the results may not be desirable. The current study reviewed the characteristics, structures, and factors affecting the performance of the WB chambers, especially the ones designed for small animal exposure to the PM. At the primary stage, the criteria and the search strategy were determined and the keywords were searched in the scientific electronic databases. Totally, 1051 articles were extracted in the first stage, and finally seven articles were adopted. The technical and design details, materials, coefficient variations (CVs) of concentration, assessment methods, type and number of laboratory animals, procedure, and animals housing conditions were extracted from the selected articles. Then the most desirable WB inhalation exposure chamber was determined based on the criteria for assessing the presented exposure chambers such as the animal housing and least CVs of the concentration in the respiratory zones of the animals under study. It was concluded that the Kimmel design was the best and the most desirable chamber structurally and geometrically, since the concentration of the particle (NaCl) injected into the chamber varied from 3.5% to 5.2%, under standard conditions.     An Examination of the Relationship between Visual Fatigue Symptoms with Flicker Value Variations in Video Display Terminal Users

    In most sensitive occupations such as nuclear, military and chemical industries closed circuit systems and visual display terminals (VDTs) are used to carefully control and assess sensitive processes. Visual fatigue is one of the factors decreasing accuracy and concentration in operators causing faulty perception. This study aimed to find out a relationship between visual fatigue symptoms (VFS) of Flicker value variations in video display terminal (VDT) operators. This cross-sectional study, conducted in 2011, aimed to examine visual fatigue and determine the relationship between its symptoms and visual flicker value changes in 248 operators of VDTs in several occupations. The materials used in this study were a visual fatigue questionnaire of VDTs and a VFM-90.1 device. Visual fatigue was measured in two stages (prior to beginning to work and 60 min later). The data were analyzed by SPSS11.5, using descriptive statistics, paired t-test, simple and multiple linear regressions, correlation and recognition coefficients. Then regression equations of changes in flicker value depending on the changes in the main domains and the changes in final score before the questionnaire were obtained. Paired t-test indicated significant differences in the mean score of visual fatigue symptoms and the mean score of flicker value between the two stages, respectively (P ≤ 0.001). Simple and multiple regressions of flicker value variations, for the last visual fatigue changes in questionnaire score and the four main domains of the questionnaire were obtained R2 = 0.851 and R2 = 0.853, respectively. Correlation coefficient in the above tests indicated reverse and significant relationships among flicker value changes with changes in questionnaire score and visual fatigue symptoms. Diagnosing the first symptoms of visual fatigue could be an appropriate warning for VDTs operators in sensitive occupations to react suitably, in behavior and management, to control or treat visual fatigue and prevent errors efficiently

    Antimicrobial Features of Cerastoderma and Didacna Double Basins Peptides

    Background: In recent years, high attention has been given to the biological activities of natural compounds and their potential antimicrobial properties. Objective: In this study, the antibacterial properties of the extracts from tissue and peptides of Cerastoderma and Didacna were studied. Materials and Methods: samples of Cerastoderma and Didacna were collected and washed. Then, the soft tissues were cut and powdered, and concentrations of 16, 8, 4, 2, 1 and 0.5 of chloroform, ethanol and methanol, and in addition extract of enzymatic hydrolysis were prepared, and their antibacterial activities against Staphylococcus aureus, Escherichia coli and Salmonella paratyphi were investigated. The disc diffusion method was used for the evaluation of strains susceptibility. Minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC) were investigated for bacterial growth inhibition. Results: Methanolic and ethanolic extracts from Cerastoderma demonstrated higher growth inhibitory effects compared to those from Didacna on E. coli and S. paratyphi and exhibited similar activities against S. aureus at concentrations 16 and 8 ug/mL. In addition, chloroform extracts of Cerastoderma and Didacna displayed similar inhibitory effects on S. paratyphi and S. aureus at concentrations 16 and 8 ug/mL which was a suitable effect, and the extract from Cerastoderma was more effective. MIC and MBC of methanolic extracts were at the lowest level, especially against S. aureus. Conclusion: It was revealed that Cerastoderma and Didacna extracts were effective as antibacterial compounds on S. aureus, E. coli and S. paratyphi species as natural agents

    Prevalence of Clinically Isolated Metallo-beta-lactamase-producing Pseudomonas aeruginosa, Coding Genes, and Possible Risk Factors in Iran

    Background & Objective: The spread of carbapenem-resistant Pseudomonas aeruginosa is a global concern. Metallo-beta-lactamase (MBL) enzymes cause extensive drug resistance among Gram-negative bacteria. The current study aimed at determining the prevalence of MBL-producing P. aeruginosa in Iran. Data extraction: A total of 43 studies were found out of which 36 were adopted. Data were collected from Google, Google Scholar, Science Direct, PubMed, Scopus, Embase, and Sciverse. The terms “Pseudomonas aeruginosa”, “metallo-beta-lactamase”, “prevalence”, “carbapenems”, and “Iran” were searched. Data from the isolates not producing MBLs were excluded from the study. Data were analyzed with Graph Pad Prism 6, meta-analysis section. Results: According to the results of the current study, 36 surveys indicated that 55% of the clinically isolated P. aeruginosa in Iran were resistant to imipenem and meropenem, among which 37.72% were the MBL producers. Among genes encoding MBLs, blaVIM and blaIMP were predominant with the prevalence of 12.91%±11.01% and 12.50%±23.56%, respectively. No report of harboring blaNDM1 and blaSPM1 by P. aeruginosa was found, similar to most of the other countries in Asia. The prevalence of blaVIM and blaIMP from burn settings were 11.50%±3.5% and 24.65%±23%, respectively. Furthermore, the prevalence of these genes was not significantly different among burn and non-burn isolates (P=0.942 and P=0.597, respectively). Moreover, no relationship was observed between the MBL production and patients’ age range. Conclusion: Approximately half of P. aeruginosa isolates were carbapenem-resistant in Iran, and approximately half were the MBL producers. The blaVIM and blaIMP were the predominant MBLs among P. aeruginosa strains, while other genes were not found in P. aeruginosa. Moreover, there was no significant difference between blaVIM and blaIMP among burn and non-burn isolates. Due to the multiple drug resistance conferred by MBLs, detection and control of their spread alongside proper therapeutic regimens in hospitals and community settings are essential to prevent infection acquisition. © 2017, IRANIAN JOURNAL OF PATHOLOGY

    Designing and validation a visual fatigue questionnaire for video display terminals operators

    Background: Along with the rapid growth of technology its related tools such as computer, monitors and video display terminals (VDTs) grow as well. Based on the studies, the most common complaint reported is of the VDT users. Methods: This study attempts to design a proper tool to assess the visual fatigue of the VDT users. First draft of the questionnaire was prepared after a thorough study on the books, papers and similar questionnaires. The validity and reliability of the questionnaire was confirmed using the content validity index (CVI) beside that of the Cronbach's Coefficient Alpha. Then, a cross-sectional study was carried out on 248 of the VDT users in different professions. A theoretical model with four categories of symptoms of visual fatigue was derived from the previous studies and questionnaires. Having used the AMOS16 software, the construct validity of the questionnaire was evaluated using the confirmatory factor analysis. The correlation co-efficiency of the internal domains was calculated using the SPSS 11.5 software. To assess the quality check index and determining the visual fatigue levels, visual fatigue of the VDT users was measured by the questionnaire and visual fatigue meter (VFM) device. Cut-off points were identified by receiver operating characteristic curves. Results: CVI and reliability co-efficiency were both equal to 0.75. Model fit indices including root mean of squared error approximation, goodness of fit index and adjusted goodness of fit index were obtained 0.026, 0.96 and 0.92 respectfully. The correlation between the results measured with the questionnaire and VFM-90.1 device was-0.87. Cut-off points of the questionnaire were 0.65, 2.36 and 3.88. The confirmed questionnaire consists of four main areas: Eye strain (4 questions), visual impairment (5 questions) and the surface impairment of the eye (3 questions) and the out of eye problems (3 questions). Conclusions: The visual fatigue questionnaire contains 15 questions and has a very remarkable validity and reliability. Usingthis questionnaire and its findings, one will be able to identify, assess and finally prevent the effective factors of VDT users' visual fatigue

    Sero-prevalence of helicobacter pylori infection in Neyshabur, Iran, during 2010-2015

    Backgrounds & Objective: The Helicobacter pylori prevalence has continuously decreased during recent years in Iran. The current study aimed at determining H. pylori prevalence in Neyshabur city, Northeast Iran, during 2010-2015. Methods: The current epidemiologic survey was conducted in Neyshabur from 2010 to 2015 to determine the prevalence of H. pylori infection. A total of 11596 participants (3681 male with the mean age of 31.7±6.2 years and 7915 female with mean age of 68.3±4.7 years) were included. The enzyme-linked immunosorbent assay kits for the detection of H. pylori and Stat Fax 3200® Microplate Reader (USA) with a sensitivity of 95% and specificity of 98% were used. Titers above 12 units were considered positive for IgG, IgA, and IgM (negative 12 U). The Chi-square t test and F test were used to analyze data. Results: The overall IgA, IgG, and IgM seropositive samples among the study participants were 852 (7.2%), 9000 (72.8%), and 1256 (5.2%), respectively. The IgA seropositivity was significantly high among the age group above 51 years, compared with the other age groups. Moreover, the IgG and IgM seropositivity were significantly high among the age groups 41 to 50 and 31 to 40 years respectively, compared with the other age groups. There was no significant difference between male and female cases regarding IgA and IgG seropositive samples, but IgM level was significantly higher among females, compared with that of the male cases. Furthermore, there was no significant alteration in IgA, IgG, and IgM seropositivity during 2010-2014 in Neyshabur. Conclusion: The prevalence of H. pylori in Neyshabur was high in the healthy population. Furthermore, the H. pylori prevalence did not change from 2010 to 2014 in the studied city. Effective approaches to improve health, educational, and socioeconomic status should be implemented to minimize and control H. pylori infection

    Molecular Typing of Klebsiella pneumoniae isolates using Repetitive Extragenic Palindromic Sequence-Based PCR in a Hospital in Tehran, Iran

    Background: The presence of extended-spectrum β-lactamases (ESBLs) is increasing worldwide and blaCTX-M1 is the predominant β-lactamase. Objective: This study was conducted to determine the ESBL production and prevalence of blaCTX-M1, blaSHV and blaTEM and AmpC genes and repetitive extragenic palindromic polymerase chain reaction (rep-PCR) pattern among Klebsiella pneumoniae isolates in Tehran from 2014 to 2016. Materials and Methods: One hundred eleven isolates were collected during the study period. The PCR was employed to detect the blaCTX-M1, blaSHV, blaTEM and AmpC genes. The genetic relation of isolates was performed using rep-PCR typing method. Results: Eighty-three and 86 isolates showed Minimum inhibitory concentration (MIC) ≥2 against ceftazidime and cefotaxime, respectively and 80 (72%) isolates exhibited ESBL production. The prevalence of blaCTX-M1, blaSHV, blaTEM and AmpC genes among ESBL producers was 92.5% (n = 74), 66.2% (n = 53), 56.2% (n = 45) and 2.5% (n = 2), respectively. The rep-PCR typing pattern of isolates showed a wide diversity, indicating the polyclonal spread of CTX-M type producing isolates. Conclusion: The findings of this study highlighted the emergence and spread of K. pneumoniae isolates producing CTX-M and other ESBL enzymes with diverse genetic backgrounds in a hospital in Tehran
